Constitutive Rhl sender (J23105+RhlI)
The Rhl sender device consists of a constitutively expressed RhlI AHL synthase (BBa_C0170) under the control of a moderate Anderson promoter (BBa_J23105). RhlI produces the C4-AHL signal that is required for activation of the RhlR transcription regulator.
Usage and biology
The Rhl sender is based on the Rhl quorum-sensing system from Pseudomonas aeruginosa. Quorum sensing is a naturally occurring mechanism that certain strains of bacteria use to regulate gene expression in response to their population density. These bacteria secrete autoinducer signalling molecules, such as N-acyl homoserine lactones (AHLs), that bind to transcription factors to alter gene expression.
The constitutively expressed RhlI AHL synthesis the C4-AHL signal that binds to RhlR to activate the transcription regulator. Activated RhlR can then bind to the RhlR-inducible promoter, pRhl, in order to initiate transcription of a downstream coding sequence.
